				<content:encoded><![CDATA[<p><b> </b>代谢组学是继基因组学和蛋白质组学之后新近发展起来的一门学科，是系统生物学的重要组成部分。基因组学和蛋白质组学分别从基因层面和蛋白质层面探寻生命的活动，而实际上细胞内许多生命活动是与代谢物相关的，如细胞信号、能量传递等都是受代谢物调控的。代谢组学正是研究在某一时刻细胞内所有代谢物的集合的一门学科。基因与蛋白质的表达紧密相连，而代谢物则更多地反映了细胞所处的环境，这与细胞的营养状态、药物和环境污染物的作用，以及其它外界因素的影响密切相关。因此有人认为，基因组学和蛋白质组学能够说明可能发生的事件，而代谢组学则反映确实已经发生了的事情。<br />
从众多的代谢产物中选取研究对象，无疑是大海捞针。代谢组学研究通过一定的手段能够帮助研究员从代谢产物海中跳出来，提供一个“航拍”的视角，一目了然地发现差异性代谢产物。然后通过已知的代谢通路逆推找出调节酶和基因，完成疾病发病机制、药物治疗机制等方面的研究。<br />

	<b>长链脂肪酸</b>
</p>
<p class="MsoNormal" style="text-indent:21pt;">
	<span>脂肪酸在生物中广泛存在，是许多复杂脂质（如甘油三酯、磷脂等）的重要成分，在细胞供氧充足的情况下，可被氧化分解为</span>CO2和H2O，为机体提供大量的能量支持。脂肪酸在信号传递中亦有重要作用，含多不饱和脂肪酸的脂质可在细胞结构中起到“脂筏”角色，利于胞内外物质的传递。脂肪酸由碳、氢、氧三种元素组成，系中性脂肪、磷脂和糖脂的主要成分。短链脂肪酸的碳链上的碳原子数小于6，系挥发性脂肪酸。中链脂肪酸的碳链上碳原子数为6-12的脂肪酸，主要是辛酸（C8）和癸酸（C10）。长链脂肪酸的碳链上碳原子数大于12。脂肪酸碳氢链一端为甲基端，一端为羧基端。通过检测游离脂肪酸水平来反映机体的脂质代谢、糖代谢及内分泌功能状态，对探索疾病的发病机理及防治有十分重要的意义。

	<span>氧化三甲胺（</span>TMAO）是一种肠源性菌群代谢物，是心血管疾病和其他慢性疾病的潜在风险因子。生物体内的胆碱、左旋肉碱、甜菜碱等在肠道菌群的作用下转化为三甲胺后在肠道中被有效吸收，经三甲胺氧化酶的作用转化为氧化三甲胺，后经肾脏代谢随尿液排出。研究发现<span>三甲胺</span>N-氧化物（TMAO）可促进动脉粥样硬化的发病，与心血管事件风险相关。TMAO成为越来越多慢性疾病的危险因素，包括肾脏疾病、肥胖、二型糖尿病和癌症。可采用LC-MS的方法检测氧化三甲胺及相关代谢物的含量变化。

	靶向代谢组学是对特定的代谢物群进行检测与分析。分析时加入了标准品的定量分析，用以对特定代谢物绝对浓度的定量检测。常规分析法有气质联用（GC-MS）和液质联用（LC-MS），我们优先利用LC-MS的方法进行代谢组分析[1]，然后通过与相同分析条件下标准品的分析结果进行比对，最终完成目标代谢群定性和定量的分析。

	针对不同靶向检测物，我们制备了多种标准品，可对 <a href="http://www.tinygene.com/metabolomics/target-metabolite/bile">胆汁酸</a>、<a href="http://www.tinygene.com/metabolomics/target-metabolite/scfa">短链脂肪酸</a>、TMAO及相关代谢物、 <a href="http://www.tinygene.com/metabolomics/target-metabolite/amino">氨基酸</a>、脂肪酸、有机酸、黄酮类、 <a href="http://www.tinygene.com/metabolomics/target-metabolite/phytohormone">植物激素</a>、 <a href="http://www.tinygene.com/metabolomics/target-metabolite/neurotransmitter">神经递质</a>等进行靶向检测。
